ABSTRACT
Introduction: Oral squamous cell carcinoma is one of the most widespread cancer which totalizes more than 90% of oral malignancies. Therefore, the conception of detaining or preventing the malignant transformation remains a viable target for future. Oxaliplatin is a third-generation platinum based chemotherapy cure that has value in the treatment against several forms of neoplasms. It forms intrastrand links between two adjoining DNA bases, hence disrupting its replication and transcription.
Aim of the study: The current work was carried out to report the oxaliplatin drug as a chemotherapeutic agent during DMBA-induced carcinoma in hamster buccal pouch, utilizing histopathology and flow cytometry analysis.
Material and methods: A total of 60 Syrian hamsters distributed as 2 animals examined for the normal pouch mucosa and 58 hamsters divided into; 6 experiments for Group I, their pouches were painted only with mineral oil. The remaining 52 animals for Group II, treated by DMBA mixed in a mineral oil. After 6 weeks, the hamsters separated into 2 subgroups; Group IIA, were persisted operated in DMBA. Group IIB, were employed to DMBA and injected with oxaliplatin.
Results: Oxaliplatin revealed effectiveness and tolerance in turn down the DMBA carcinogenesis procedure. Additionally, the chemotherapeutic results of oxaliplatin detected a significant reduction relation to the DNA aneuploidy and the S-phase fraction throughout the tumorigenic activity.
Conclusion: Oxaliplatin provided a proper strategy as a chemotherapeutic curing for control oral carcinogenesis process with a notable reduction of cancer incidence through reducing the nuclear proliferation activity and induction of cellular apoptosis.
ABSTRACT
Objectives: The aim of the present study is to evaluate CD44 expression to detect cancer stem cell activity along with analyzing the DNA ploidy, to predict the activity of oral carcinogenesis.
Materials and method: Forty formalin fixed paraffin embedded tissues of oral lesions were used and divided equally into 4 groups; different premalignant white lesions, well, moderate, and poorly differentiated squamous cell carcinomas. All specimens were immunostained using CD44 antibodies. Moreover, the same paraffin blocks for each case used to prove the differentiation in DNA ploidy between groups using flow cytometer.
Results: The CD44 immunoexpression showed a marked statistical significant difference between premalignant white lesions and poorly differentiated squamous cell carcinomas. Flow cytometric analysis of DNA parameters between the tested groups revealed a powerful difference of DNA ploidy between the premalignant group and other malignant groups. The S-phase fraction showed significant difference between poorly differentiated OSCC and premalignant lesions with no effective differences between the malignant lesions.
Conclusions: The findings of the present study suggest that the CD44 immunoexpression collaborates with the DNA content analysis presented a real indicator in prediction the behavior of oral premalignant and malignant lesions.
Abstract
Introduction: Oral squamous cell carcinoma (OSCC) management is challenging due to high tendency of local invasion and metastasis. Cancer stem cells hold high significance as they have self-renewal ability, which further allows cancer progression and metastasis. Hence, it is crucial to evaluate different specialised markers for stem cells, such as CD44, to detect their role in tumour metastasis. Flow cytometry (FCM) offers a quick and automated assessment of ploidy status and cell proliferation of the neoplasm by resolving the nuclear DNA contents.
Aim of the study: The objective of this paper is to evaluate the CD44 expression and analyse DNA content by FCM to predict the expansion of lymph node metastasis in patients with OSCC.
Material and Methods: 50paraffin-embedded tissues of metastatic and non-metastatic lymph node OSCC were immuno stained by CD44 for assessing cancer stem cell activity in each lesion. Furthermore, each selected tissue underwent flow cytometric analysis to demonstrate the DNA activity between the tested groups.
Results: The CD44 expression in OSCCs showed a marked difference between the metastatic and non-metastatic lymph node cases. Furthermore, flow cytometric analysis of the DNA parameters between the tested groups revealed a powerful difference of DNA ploidy. The S-phase fraction (SPF) between the groups showed no compelling result. All specimens had a higher CD44 expression, aneuploid DNA content and high SPF, which demonstrated deposits of cervical metastatic lymph nodes.
Conclusions: The CD44 and the flow cytometric analysis of the DNA ploidy correlation offer a significant prediction to determine the OSCC competence.
ABSTRACT
Objective: This trial preformed to evaluate clinically and radiographically, the efficacy of growth factors producing/inducing materials; Advanced-Platelet-Rich Fibrin (A-PRF) and Metformin (MF) respectively, in the surgical treatment of intrabony periodontal defects.
Methods: Forty-eight systemically healthy chronic periodontitis patients with intrabony defect were divided equally into 4 groups. First Group, patients treated by open flap debridement (OFD). Second Group, patients treated by OFD + 1% MF gel. Third Group, patients treated by A-PRF, which inserted into the intra bony defect (IBD). Fourth Group treated by (1% MF + A-PRF). Parameters were gathered at baseline 6 and 9 months.
Results: The reduction in probing depth (PD) and clinical attachment level (CAL) was greater in the A-PRF + 1% MF patients group than other groups. Combination of A-PRF + 1% MF showed statistically significant reduction of IBD greater than all other groups.
Conclusions: Usage of combination of A-PRF + 1% MF seems to be superior in gaining bone than surgical treatment by OFD, OFD + A-PRF or OFD + MF only
